Viagra treats kids' deadly lung condition

Impotence drug helps children with pulmonary arterial hypertension breathe easier and walk farther

A small pilot study suggests the impotence drug Viagra can improve breathing and exercise capacity in children with an often fatal lung condition called pulmonary arterial hypertension (PAH).
PAH is a condition in which blood pressure in the arteries that supply the lungs is extremely high. If the pressure soars high enough, the heart can't keep up, causing fatigue, dizziness, shortness of breath and eventually heart failure and death. The condition can be inherited, caused by a congenital heart defect or result from another chronic heart or lung disease.

"Untreated, children usually die within one year of diagnosis," says Dr. Ian Adatia, an associate professor of pediatrics at the University of California in San Francisco, in a statement. He says few children live five years past diagnosis even with the current best therapy, a drug called prostacyclin that is inconvenient to administer and comes with side-effects such as jaw and muscle pain and facial flushing.

When Adatia was in his former position as head of the pulmonary hypertension clinic at the Hospital for Sick Children in Toronto, he and his colleagues decided to test an alternative treatment known more for improving blood flow in the penis than in the lungs -- Viagra, also known as sildenafil.

The researchers gave Viagra pills to 14 children with PAH for one year. After the treatment, the children could walk an average of 154 metres farther during a six-minute period. "That's an appreciable increase that compares favourably with the 44- to 70-metre increases seen with other pulmonary arterial hypertension drugs," Adatia says.

Children could also breathe more easily by the end of the study, and experienced few side-effects.

All children survived the study period, although one died after 22 months. In comparison, other children with PAH have a survival rate of only 37 per cent after one year.

The study was funded by Pfizer, Inc., the makers of Viagra.

CoTherix Announces Fourth Quarter Ventavis Sales and 2006 Revenue Guidance

Ventavis Sales Increase 30 Percent Over Previous Quarter

SOUTH SAN FRANCISCO, Calif., Jan. 19 /PRNewswire-FirstCall/ -- CoTherix, Inc. (Nasdaq: CTRX - News) today announced that preliminary net product sales of Ventavis®(iloprost) Inhalation Solution in the fourth quarter of 2005 totaled approximately $11.0 million, up 30 percent from $8.4 million in the third quarter of 2005. Preliminary 2005 net product sales for Ventavis, which was launched in March 2005, were approximately $23.9 million. Ventavis is indicated for the treatment of pulmonary arterial hypertension (PAH) in patients with New York Heart Association Class III or IV symptoms (World Health Organization Group I).
The company currently estimates that Ventavis net product sales in 2006 will be between $65 million and $70 million. CoTherix plans to provide additional financial guidance for 2006 on March 8, 2006 when the company reports its full fourth quarter and year end financial results.

"Ventavis, as the first inhaled prostacyclin therapy, has established an important role in PAH treatment. Since launch, Ventavis has become more convenient for patients with the introduction of the new, portable I-neb inhalation device in mid-December. In less than one month, approximately ninety percent of patients were converted to the I-neb device," said Donald J. Santel, Chief Executive Officer of CoTherix, Inc. "We believe the added convenience of the I-neb device will provide further momentum for strong sales growth for Ventavis in 2006."

Santel added, "With our existing capital and projected revenue growth, we believe we have the resources necessary to maximize the Ventavis opportunity by continuing to invest in further clinical development. In addition, during the first half of 2006, we plan to initiate the REVEAL Registry to contribute to a better understanding of PAH disease management and improving patient care."
